Media by purpose
- General purpose — TSA, nutrient agar, brain heart infusion. Broad recovery, no selection.
- Selective — MacConkey, XLD, Baird-Parker. Inhibit unwanted flora to isolate targets.
- Differential — blood agar, EMB. Distinguish organisms by colony appearance.
- Chromogenic — enzyme-specific substrates give colour-coded identification, cutting confirmation steps.
- Enrichment broths — recover stressed or low-count organisms before plating.
Preparation and storage
Dehydrated media are hygroscopic. Keep containers tightly closed in a cool, dry place and record the date first opened — caking, colour change or hardening indicates the powder has absorbed moisture and should not be used.
Follow the label for autoclave time and temperature. Over-autoclaving darkens media and degrades selective agents; heat-sensitive supplements are added after cooling to about 45-50 °C.
Quality control and documentation
Each lot ships with a certificate of analysis covering growth promotion, selectivity, pH and physical appearance against reference strains.
Laboratories operating under ISO 17025, CLIA or USP will find the lot documentation sufficient for their QC records. Certificates are available before purchase on request.
