The Milenia Biotec HybriDetect 2T (MGHD 2) is a sophisticated "open platform" lateral flow dipstick designed for the simultaneous detection of two different analytes within a single sample. Unlike standard single-target strips, the HybriDetect 2T features two distinct test lines and one control line, enabling researchers to perform multiplex assays or include essential internal amplification controls (IAC). This tool is ideal for developing rapid, qualitative, or semi-quantitative tests for nucleic acids, proteins, and antibodies without the need for complex equipment.

The Multiplex Detection Principle
The HybriDetect 2T system relies on a specific three-label strategy to differentiate between two targets on the same strip. Gold nanoparticles conjugated with anti-FITC antibodies serve as the universal reporter. The separation occurs at the test lines:
- Test Band 1 (T1): Contains Streptavidin to capture Biotin-labeled analytes.
- Test Band 2 (T2): Contains Anti-Digoxigenin antibodies to capture Digoxigenin (DIG)-labeled analytes.
- Control Band (C): Ensures the test runs correctly.
This configuration requires one target to be labeled with FITC+Biotin and the second target with FITC+DIG, as shown below:

1. Multiplex Genomic Detection (DNA/RNA)
In molecular diagnostics, the HybriDetect 2T is widely used to detect two different gene amplicons simultaneously (e.g., Pathogen A and Pathogen B) or to confirm a negative result using an Internal Amplification Control (IAC). One amplicon is generated with a Biotin-labeled primer, and the other with a Digoxigenin-labeled primer. Both must use a FITC-labeled probe or primer for visualization.

2. Multiplex Protein Detection
The platform is equally powerful for developing multiplex immunoassays. Researchers can detect two distinct proteins (Protein A and Protein B) in a single run. This is achieved by using two different "sandwich" pairs: one utilizing a Biotinylated capture antibody and the other a Digoxigenin-labeled capture antibody. Both detection antibodies are conjugated to FITC to bind the gold particles.

3. Multiplex Antibody Detection
For serological applications, the HybriDetect 2T allows for the differentiation of antibody classes (e.g., IgX vs. IgZ) or specificities against different antigens. By immobilizing specific capture molecules labeled with Biotin and Digoxigenin, users can screen for multiple immune responses on one strip.
