Activin A is a key member of the TGF-β superfamily, essential for proper development and produced by various cell types. It functions as a bioactive disulfide-linked homodimer, binding to Activin receptors (RIA, RIB, RIIA, RIIB) to activate SMAD signaling pathways. This signaling regulates processes such as stem cell maintenance, differentiation, B-cell proliferation, wound healing, metabolism, and gonadal development.
It is synthesized as a precursor, then cleaved to form a mature A chain, which dimerizes to become active. Its activity is tightly regulated by decoy receptors, secreted inhibitors (e.g., Follistatin), and alternative subunit incorporation. In research, recombinant Activin A is widely used to support stem cell proliferation and drive differentiation into endodermal and pancreatic lineages.
