Manual kits or automated extraction?

Spin-column kits are economical and flexible — ideal below roughly 20 samples per run and for varied sample types.

Automated magnetic-bead extractors pay for themselves when you process consistent batches daily. They remove hands-on time, eliminate operator-to-operator variability, and reduce cross-contamination risk. For diagnostic laboratories running the same panel every day, automation is usually the right call.

Matching chemistry to sample type
  • Blood and swabs — standard viral or genomic kits handle most clinical specimens
  • Tissue — requires proteinase K digestion and often mechanical homogenization first
  • FFPE — needs deparaffinization and extended digestion; expect fragmented yield
  • Plant and soil — high in inhibitors, requires dedicated chemistry
  • Bacteria and fungi — cell wall lysis step is essential, especially for Gram-positive organisms
Inhibitors and downstream failure

Most PCR failures trace back to extraction, not amplification. Heme from blood, humic acid from soil, polysaccharides from plants and residual ethanol from the wash step all inhibit polymerase. If your assay fails on a specific sample type, check purity ratios and consider a chemistry designed for that matrix before troubleshooting the PCR.

Yield, purity and storage

A 260/280 ratio near 1.8 indicates clean DNA; near 2.0 indicates clean RNA. A low 260/230 ratio signals guanidine or ethanol carry-over. Store DNA in elution buffer rather than water — the slight buffering protects against acid hydrolysis. RNA should go to −80 °C promptly and avoid repeated freeze-thaw.
