Most PCR failures trace back to extraction, not amplification. Heme from blood, humic acid from soil, polysaccharides from plants and residual ethanol from the wash step all inhibit polymerase. If your assay fails on a specific sample type, check purity ratios and consider a chemistry designed for that matrix before troubleshooting the PCR.
A 260/280 ratio near 1.8 indicates clean DNA; near 2.0 indicates clean RNA. A low 260/230 ratio signals guanidine or ethanol carry-over. Store DNA in elution buffer rather than water — the slight buffering protects against acid hydrolysis. RNA should go to −80 °C promptly and avoid repeated freeze-thaw.
