Choose the volume range, not the maximum

Air-displacement pipettes are least accurate at the bottom of their range. Dispensing 20 µL with a 200 µL pipette introduces noticeably more error than using a 20 µL instrument.

As a rule, work in the upper half of the nominal range. A lab handling 1-10 µL routinely needs a dedicated 10 µL pipette, not a 100 µL unit turned down.

Calibration and drift

Pipettes drift. Seals wear, springs fatigue, and a dropped instrument can shift calibration invisibly. ISO 8655 recommends verification at least annually, and every six months for pipettes in daily use or in regulated environments.

A simple gravimetric check with an analytical balance catches most problems between formal services. If your assay CVs are creeping up, check the pipettes before blaming the reagents.

Multichannel and plate work

Eight- and twelve-channel pipettes transform 96-well plate work, but tip fit becomes critical — a single loose tip on one channel produces a column of bad data that is easy to miss. Verify all channels seat evenly, and check channel-to-channel consistency periodically, not just total volume.

Repeating and positive displacement

Repeating pipettes dispense many aliquots from one aspiration, cutting time and strain during plate filling. Positive displacement is the answer for viscous, volatile or dense liquids — glycerol, DMSO, organic solvents — where air-displacement pipettes under-deliver because the air cushion compresses or the liquid evaporates into it.
