Premixed or prepare in-house?

Preparing buffers in-house is cheaper per litre but costs technician time, requires calibrated balances and pH meters, and introduces lot-to-lot variability that is difficult to trace when an assay drifts.

Premixed buffers arrive verified for pH, conductivity and nuclease activity. For regulated laboratories and any workflow where a failed run costs more than the reagent, premixed is usually the better economics.

Water grades

Not all purified water is interchangeable. Type I ultrapure (18.2 MΩ·cm) is required for molecular biology, HPLC and trace analysis. Type II serves general laboratory use, buffer preparation and media.

For nucleic acid work, confirm the water is certified DNase- and RNase-free. Deionization alone does not remove nucleases — that requires DEPC treatment or validated filtration.

Agarose and acrylamide grades

Standard agarose covers routine DNA electrophoresis. Low-melting-point agarose allows gel extraction and in-gel enzymatic reactions. High-resolution agarose separates small fragments where standard grades smear.

Acrylamide is a potent neurotoxin in its unpolymerized form. Handle solutions and powder with gloves in a ventilated enclosure, and prefer premixed solutions over weighing powder whenever possible.

Storage and shelf life

Most buffers store at room temperature but some components precipitate when cold — check the label before refrigerating. Record the date opened on every bottle. Buffers containing reducing agents such as DTT or beta-mercaptoethanol degrade once opened and should be dated and replaced on schedule rather than used until empty.
