Plasma is a goldmine of biological insight, but also one of the most challenging fluids to analyze. Highly abundant proteins such as human serum albumin (HSA) and immunoglobulin G (IgG) make up nearly 70 % of total plasma protein content, masking the low-abundance biomarkers that matter most.
In just 10 minutes, ProtiPrep™ Pop2 removes over 97 % of HSA and IgG from only 10 µL of human plasma, leaving the sample ready for downstream proteomic analysis. A patented centrifugal-flow microspin device is combined with prefilled buffer compartments in the lid, so the complete depletion workflow takes place in a single tube — no buffer preparation, no column conditioning, no transfer steps.

Performance Data
- Depletion efficiency (ELISA): 97.27 % HSA and 98.62 % IgG. Pooled plasma from 134 donors, n = 61 samples processed in three technical replicates, measured with the HSA ELISA Kit (KE00076, Proteintech) and the Human IgG Total ELISA Kit (Thermo Fisher).
- Proteome depth (LC-MS/MS): approximately 100 additional distinct protein groups identified in depleted samples (n = 9) versus undepleted neat plasma (n = 3) — 327 + 103 versus 328 proteins. Acquired on an Orbitrap Exploris 480 (Thermo Fisher Scientific), analyzed with R 4.5.2 and SpectroPipeR 0.4.6.1, counting only proteins identified with at least two unique peptides. Data generated by ProTec Diagnostic GmbH, Greifswald, Germany.
- SDS-PAGE: strong reduction of the HSA (66.5 kDa) and IgG heavy-chain (50 kDa) and light-chain (25 kDa) bands in depleted samples, with specific capture confirmed in the bead fraction (Novex™ WedgeWell™ 4–20 % Tris-Glycine gel).
Workflow
- Add sample (10 µL plasma)
- Screw the lid on to release the prefilled buffer
- Tap 10×
- Shake for 10 seconds
- Incubate 5 minutes
- Centrifuge
- Take out the column
- Collect the sample — ready for downstream analysis
