Upright or inverted?

Upright microscopes view specimens from above — slides, smears, histology sections, blood films. The standard for clinical and teaching work.

Inverted microscopes view from below, so you can look through the bottom of a culture flask or plate without opening it. Essential for live cell culture, and the only practical option for monitoring confluence.

Contrast methods and what they reveal
  • Brightfield — stained specimens. Requires contrast from the sample itself.
  • Phase contrast — unstained living cells. The default for culture work.
  • Darkfield — scattering objects on a black background. Spirochetes, diatoms, particulates.
  • Fluorescence — labelled targets. Requires matched filter cubes for your fluorophores.
  • Polarizing — birefringent material. Crystals, fibres, and synovial fluid analysis for gout.
Objectives determine the result

The objective, not the body, sets image quality. Plan objectives correct field curvature so the whole field is in focus — important for photography and screen viewing. Semi-apochromat and apochromat corrections reduce colour fringing progressively. Numerical aperture, not magnification, determines resolution: a 40x objective with NA 0.65 resolves finer detail than a 40x with NA 0.45.

Documentation and teaching

Trinocular heads accept cameras for image capture and are worth specifying if you publish, archive or share findings. For teaching, dual-view heads and screen-based digital microscopes let an instructor and student view the same field simultaneously.
